西藏淡红蜡伞菌多糖的提取及单糖组成研究

Study on Extraction of Polyoses and Compositions of Monosaccharide of Hygrophorus Russula in Tibet

采用正交试验优化西藏淡红蜡伞菌粗多糖提取工艺条件,通过蒽酮-硫酸法比色测定提取粗多糖中的总糖含量,并应用气相色谱法对淡红蜡伞菌多糖中单糖的组成进行分析。结果表明,西藏淡红蜡伞菌多糖最佳提取条件是提取时间4h,温度80℃,料水比1:10,乙醇沉淀浓度85%,多糖得率为6.52%;比色测定粗多糖中的多糖含量在49.17%～69.86%。色谱分析得出该多糖是由5种单糖组成的杂多糖。

The extraction technological conditions of polyoses of the hygrophorus russula in Tibet were designed using orthogonal methods. The total sugar contents in the polyoses were assayed by anthrone - sulfuric acid colorimetric estimation, and the compositions of monosaccharide in polyoses which belonged to hygrophorus russula were analysed by employing Gas Phase Chromatography. The results showed as follows： The optimum extraction time was 4h; the temperature was 80℃ ; the raio of material and water was 1 ： 10 ; the deposit concentration was 85 % ; the yield of polyoses was at the range of 49.17- 69.86% by colorimetric estimation. The polyoses were composed by 5 monosaccharide using Gas Phase Chromatography.
