摘要
目的探讨糖网一号对糖尿病大鼠视网膜细胞间粘附分子-1(ICAM-1)表达的影响及其保护作用。方法将造模成功的大鼠随机分为糖尿病模型组、二甲双胍组、糖网一号小剂量组、糖网一号大剂量组,每组12只,并设正常对照组(CON),相应处理各组大鼠。造模后及6个月后,分别测血糖及血液流变指标,并取大鼠眼球的石蜡切片进行免疫组织化学检测,对视网膜细胞间粘附分子-1(ICAM-1)的表达灰度进行计算机图像半定量测定。结果糖网一号组和二甲双胍组的血糖和血液流变学检测结果相比,前者优于后者,而且糖网一号大剂量组的疗效优于小剂量用药。糖网一号大剂量组ICAM-1表达灰度值较模型组显著降低,接近正常组,而二甲双胍组及糖尿病小剂量组则介于二者间。结论中药复方糖网一号能够有效降低血糖,改善血流变,抑制糖尿病大鼠视网膜ICAM-1的表达。
OBJECTIVE To investigate the influence of Tangwangyihao(糖网一号, TWYHD)on the experssion of intercellular adhesion molecular - 1 ( ICAM - 1 ) in the streptozoein (STZ) - induced diabetes mellitus (DM) rat' s retina.METHODS The rats with diabetic were divided randomly into 4 groups: diabetes model group(M), Mefformin Hy- droehloride group(A), small dose preparation group of TWYHD(B), and large dose preparation group(C). Normal control group(CON) was also set. Sections were taken from the rat's eyes after 6 months, retinal frozen and paraffin sections were stained by immunhistoehemical SP and HE method respectively and analyzed by computer - picture analysis system. RESULTS There was an obviously coacervation and viscousness status of the diabetic rats. Retinal changes and all of the parameters which related to the blood rheology were much better in the treated group with TWYHD than in the model group, and the treated groups with medicine, such as B and C, were better than A, especially C was very similar with the CON(normal group) . CONCLUSIONS Early treatment with TWYHD,may be helpful in effectively controlling the expression of ICAM - 1. And may also have good curative effect on diabetic microvascular complications.
