摘要
进行了苏铁种子、子叶、鳞片、顶芽的试管育苗和多头苏铁诱导的研究。结果表明:试管播种的种子比常规播种提早发芽6个月,发芽率高;在ER+BA2mg/L+IBA0.4mg/L+苏铁外种皮汁2mL/L培养基中,多头诱导率达76.7%。离体培养的子叶和鳞片的芽诱导率分别达到53.3%和33.3%;试管苗在4~7月份移栽为好,成活率可达90%以上。
The plantlet regeneration, with multi-shoots especially, derived from seed, cotyledon, squama and tip-bud of Cycas revolute Thunb. was studied in vitro. The results showed that the germination of seed cultured in vitro could be six months earlier than that of sowing in normal condition. When inoculated on ER + BA 2 mg/L + IBA 0. 4 mg/L + juice of Cycas revolute Thunb. episperm 2 mL/L, the rate of multi-shoots was 76. 7%. The differentiation rate of bud derived from cotyledon and squama was 53.3% and 33.3% , respectively. It will be better when plantlets were transplanted from April to July; and the survival rate was more than 90%.
