摘要
目的从细胞凋亡角度探讨低温、辐射、中药综合治疗大肠癌的机制。方法采用Hoechst33258荧光染色技术,检测低温、紫外线两种物理因素及中药苦参碱分别及联合运用时的Colo320细胞凋亡率。结果倒置显微镜下均可以观察到细胞凋亡情况。经单纯低温-4℃,0℃,4℃处理,凋亡率为30.98%、31.72%、33.06%;经单纯15W紫外灯下50、100、150cm处照射,凋亡率为35.56%,22.69%,15.08%;经低温与紫外照射共同影响,其凋亡率为42.31%;经低温联合紫外照射及苦参碱共同作用,凋亡率达48.17%。结论Co-lo320细胞凋亡反应在低温、紫外照射及苦参碱的共同影响下,其诱导凋亡作用明显提高。
Objective: To research the mechanism of low temperature, irradiation and Chinese traditional medicine treating the colorectal cancer from the perspective of apoptosis. Methods: Hoechst 33258 fluorescent staining detected the colo320 tumour cell' s respective and jointed apoptosis rate by two physical factors of low temperature and ultraviolet irradiation combining matrine. Results: Each apoptosis situation could be inspected through the inverted microscope. After the low temperature about -4℃ , 0℃ and 4℃ , the apoptosis rates were 30. 98%, 31.72%, 33.06%. After the 15W ultraviolet irradiation in the 50, 100, 150cm position, the apoptosis rates were 35.56%, 22.69%, 15.08%. After low temperature and ultraviolet irradiation, it was 42.31% ; After low temperature unite ultraviolet irradiation and the matrine, it reached 48. 17% Conclusion: The Colo320 cell' s apoptosis rate is significantly increasing responds to low temperature unite uhraviolet irradiation and matfine.
