摘要
目的探讨缺氧条件下缺氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)对人舌鳞癌细胞株 Tca8113细胞血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法采用 RT-PCR 技术半定量检测人 Tca8113细胞常氧和缺氧培养0.5、1、3、6、12和24 h HIF-1α、VEGF 基因的表达。结果缺氧条件下 HIF-1α基因的表达呈先升后降的趋势,在缺氧6、12 h 表达显著高于常氧组(P<0.05);VEGF 的表达在缺氧培养0.5、1、3、12及24 h 均高于常氧组(P<0.05);VEGF 基因的表达在缺氧初始阶段随 HIF-1α基因表达的增强而升高,但 Spearman 相关分析显示两者在缺氧24 h 内相关性不明显(r_s=0.5750,P>0.05)。结论缺氧条件下 Tca8113细胞中 VEGF 的高表达受 HIF-1 α的调控和影响,是多种因素作用的结果。
Objective To investigate the effect of hypoxia inducible factor-1α (HIF-1α) on vascular endothelial growth factor (VEGF) expression in Tca8113 cells under hypoxia. Methods The expression of the mRNA of HIF-1α and VEGF in Tca8113 ceils was examined by RT-PCR technique at different culture times(1/2 h,1 h,3 h,6 h,12 h,24 h) under normoxic and hypoxic conditions. Results The expression of HIF-1α under hypoxia showed the trend of increasing first and then decreasing, and was higher than that of the control ( normoxic group) at 6h and 12 h ( P 〈 0.05 ). The expression of VEGF under hypoxia was higher than that of the control group at 1/2 h, 1 h,3 h, 12 h,24 h (P 〈 0.05 ). The expression of hypoxia-induced VEGF mRNA increased with the increased expression of HIF-1α mRNA in the cell lines tested at the initial stage of hypoxia. But no statistical significant association was observed between HIF-1α and VEGF expression within 24 h under hypoxia ( rs = 0. 5750, P 〉 0. 05 ). Conclusions The increased expression of VEGF in Tca8113 cells might be mediated by multiple factors, including HIF-1α.
