摘要
目的探讨标准化G422胶质瘤动物模型肿瘤细胞的生长特点。方法将接种G422胶质瘤细胞接种于国际标准化小鼠双侧上肢皮下,每侧分别注射0.1、0.2、0.3ml。接种后5、10、15、20d观察小鼠的生存状态及肿瘤的生长特性;用HE染色及胶质纤维酸性蛋白(GFAP)和S-100蛋白免疫组化分析;AnnexinV-PI观察肿瘤细胞的凋亡情况。结果标准化G422胶质瘤动物模型生长稳定;GFAP及S-100蛋白免疫组化表达强阳性,符合胶质源性肿瘤的特点;肿瘤细胞DNA呈多倍体峰,且早期出现凋亡峰。结论标准化G422胶质瘤动物模型,生长特征及病理表现与人脑胶质母细胞瘤相似,是一种理想的动物模型,为进一步研究提供了重要的平台。
Objective To explore the standard rat G422 glioblastoma multiform model. Methods The standard rats( Institute of Caneer Research rat, ICR rat) were inoeulated with the tumor of G422 glioblastoma multiform eell line. The ICR rats were injeeted with 0.1 ml,0.2 ml,0.3 ml tumor liquid. After 5,10,15 and 20 days,the living eondition and the eharaeter of tumor were observed. Tumor was eonfirmed with HE and staining of glial fibrillary aeidie protein (GFAP) and S - 100 protein immunohistoehemistry and the apoptosis was observed by Annexin V - PI staining. Results The standard rat G422 glioblastoma multiform model reliant and high rate of tumor produetion. The GFAP and S - 100 protein were obviously positive. The apoptosis was oeeurred in early stage. All these demonstrated this model resembled histopathologieal features of human glioma. Conclusions The international ICR rat G422 glioblastoma multiform model resembles histopathologieal features of human glioma,whieh ean be used as a perfeet model to study the eftology of glioblastoma multiform, It eould beeome an important tool for further research.
