液相色谱/质谱联用测定元胡止痛软胶囊中延胡索乙素的含量

Determination of Tetrahydropalmatine in Yuanhuzhitong Soft Capsule by LC-MS

目的:建立灵敏、准确的液相色谱/质谱法测定元胡止痛软胶囊中延胡索乙素的含量。方法:样品经甲醇提取后,采用Lichrospher^(TM)C.18柱(250mm×2.1mm,5μm)分离,甲醇-20mmol·L^(-1)乙酸铵溶液(70:30)为流动相。质谱采用电喷雾离子化(ESI)方式,正离子采集,以选择离子(SIR)方式进行定量测定。结果:延胡索乙素的的线性范围为2.0～10.0ng·ml^(-1),平均加样回收率为98.1%,RSD为0.78%。结论:方法简便、灵敏、准确,可用于元胡止痛软胶囊中延胡索乙素的含量测定。

Objective： To establish a method for determine the content of tetrahydropalmatine in Yuanhuzhitong soft capsule by LC/MS. Method： Sample was extracted by mathanol, Lichrospher^TM C-18 column（250 mm × 2.1 mm,5 μm）was used, mobile phase was mathanol 20 mmol·L^-1 -ammonium acetate （ 70： 30）. Mass spectrometry electrospray ionization model （ ESI^＋ ）, and SIR function were used for quantification. Result： The linear range of tetrahydropalmatine was 2.0 - 10.0 ng·ml^-1 The everage recovery was 98.1% with a RSD of 0.8% （ n =6）. Conclusion： The method is simple, sensitive and accurate, it can be used for determination the content of tetrabydropalmative in yanhuzhitong soft capsules.
