水杨酸钠对幼年豚鼠螺旋神经节GAD蛋白表达和ABR阈值的影响

摘要 目的:观察水杨酸钠给药对幼年豚鼠螺旋神经节谷氨酸脱羧酶(GAD)蛋白表达和听性脑干诱发电位(ABR)阈值的影响。方法:将48只出生7d的幼年健康豚鼠分为4组,每组12只。A组:对照组;B组:低剂量水杨酸钠给药组(200mg/kg.d-1);C组:中剂量水杨酸钠给药组(300mg/kg.d-1);D组:高剂量水杨酸钠给药组(450mg/kg.d-1)。给药前和给药15d后检测ABR阈值,然后处死动物采用免疫组织化学染色方法检测豚鼠螺旋神经节GAD蛋白表达。结果:①给药15d后B、C、D3组ABR阈值较对照组均有明显提高(P<0.05),B、C、D3组以D组ABR阈值提高最为明显,C组次之,B组ABR阈值改变最小;②B、C、D3组GAD蛋白表达积分光密度值较对照组显著降低(P<0.01);B、C、D3组以D组GAD蛋白表达积分光密度值降低最为明显,C组次之,B组GAD蛋白表达改变最小。结论:水杨酸钠可显著提高豚鼠ABR阈值并使豚鼠螺旋神经节GAD蛋白表达降低,其改变豚鼠螺旋神经节GAD蛋白表达以及ABR阈值与给药剂量有关。 Objective:To study the effects of sodium salicylate on the expression of GAD protein and ABR threshold in spiral ganglion of juvenile guinea pigs. Methods:Forty-eight guinea pigs were divided into four groups (Group A; Group B; Group C; Group D); ABR was detected before administration and after administration for 15 days; The protein expression of GAD was measured by the method of immunohistochemistry. Result:(1)ABR Threshold of sodium salicylate groups (Group B; Group C; Group D) was increased remarkably compared with the control after administration for 15 days. ABR Threshold of Group D was the highest among the three groups of administration of sodium salicylate while that of Group B was the lowest. (2)The protein expression of GAD of sodium salicylate groups was decreased compared with that of the control after administration for 15 days. The protein expression of Group D displayed the most visible degression among the three administrations. Conclusion:The results suggest that administration of sodium salicylate decreased markedly the protein expression of GAD and increased remarkably ABR Threshold of the sodium salicylate groups and these changes were related to the dosage of sodium salicylate.
