联合化疗敏感性测定后瘤细胞核的形态观察及DNA定量分析

Nuclear Morpholgy and DNA Content during Testing Chemosensitivity

应用口腔颌面部恶性肿瘤ＣＴＸ免疫抑制鼠ＳＲＣ移植瘤，经ＰＭ、ＰＶＦ、ＣＶＰ治疗后结合癌细胞形态的改变、Ａｇ—ＮＯＲ及计算机自动图象分析仪测定判断药物的敏感性。结果显示：①形态学观察治疗后癌细胞呈明显退行性改变，核肿胀，核边界不清，核染色质颗粒模糊，核有固缩。未化疗对照组，瘤细胞生长活跃核分裂相多见，排列密集。②Ａｇ—ＮＯＲ平均计数与细胞增殖活性，核仁融合功能，细胞核倍体数等有关。化疗敏感组Ａｇ—ＮＯＲ平均计数明显减少。③Ｆｅｕｌｇｅｎ染色图象分析检测，化疗敏感组ＤＮＡ指数明显偏低。提示联合化疗敏感组严重抑制细胞ＤＮＡ合成。

An experimental three combined chemotherapy study on a transplantable maglignant tumor in oral maxillofacial region in CTX immunosppress mice subrenal capsule assay was undertaden.The results showed that:(1)The tumor cells retrogressed markedly after PM.PVF.CVP combined chemothrapy.(2)Granules of Ag-NOR in the groups of chemosensitivity and the control groups had significant differeces.The mean number of Ag-NOR profoundly reduced in the groups of chemosensifivity.(3)DNA content analysis demonstreated that the groups of chemosensitivity DNA index profoundly reduced.It suggested that the synthesis of DNA in the groups chmosensitivity could be inhibited.
