摘要 目的研究人B淋巴母细胞株Yam1B产生的免疫调节团子Yam-SF的免疫调节作用。方法取Yam1B细胞培养上清,于体外培养体系中观察其对人T细胞及B细胞增殖、免疫球蛋白产生、IL-2产生及IL-2表达的影响;测定分子量,并研究其理化性质;观察抗TGF-beta抗体对其生物活性的影响。结果Yam-SF可抑制人T细胞及B细胞的增殖,并抑制正常B细胞免疫球蛋白的产生,但对PHA刺激的人淋巴细胞兄-2的产生以及IL-2受体(IL-2R)的表达无抑制作用。Yam-SF经56℃处理10min或在pH10.0的环境中处理6h抑制活性丧失,Yam-SF经pronase水解消化而失活,在pH2.0的环境中保持其抑制活性。凝胶过滤实验表明,培养上清抑制活性分布在分子量43kD~67kD范围内,且抗TGF-beta抗体不能阻断其抑制活性。结论Yam-SF是一种与TGF—beta等免疫抑制因子不同的新的免疫调节团子。 To investigate the immunoroplatory activity of Yam-SF which was spontanansly pro-duced by Yam 1B, a huxnan B lymphoblastord .lineMethods To add Yam 1B culture supamatant into the human lymphocyte culture systern and check the proiferation of human T and B lymphocytes and aseay the immunoglobulin generation by normal B cells or IL -2 preduction and the mpression of 1L-2R. The physical and chemical characters and the effect of anti -TGF-beta antibody on the immunoregulatory ac-tivity of Yam-SF were investigted.Results Ywm-SF suppressed the proihation of human T and B lymphocytes and inhibites im-munoglobulin generation by normal B cells, but did not inhibit neither the expression of 1L-2 receptor nor the preduction of IL-2 in the lymphocytes stimulated with PHA. The inhibitory activity of Yam-SF was inactivated at 56℃ 10 min and at pH10. 0 for 6h and abrogated by digestion with pronase, but resistant to pH2. 0 a dministration. Gel-filtration column as-say showed a peak of inhibitory activity of cell proif-eration associated with molecules of apparent MW of 43kD to 67kD. The inLhibitory activity was notblocked by the anti-TGF-beta antidriy.Condusion The Yam-SF appeared to represent a nove immunoregulatory factor which differ frim TGF-beta and other irnrnne inhibited factors.
