普伐他汀对醛固酮诱导心脏成纤维细胞内皮素A型受体mRNA影响 被引量:3

摘要 目的:观察普伐他汀对醛固酮诱导新生大鼠心脏成纤维细胞内皮素A型受体(ET-AR)mRNA表达的影响及其机制。方法:采用胰酶消化法和差速贴壁分离法获取新生大鼠心脏成纤维细胞,应用RT- PCR的方法测定ET-AR mRNA的表达。结果:醛固酮(0.1μmol·L^(-1))可诱导心脏成纤维细胞中ET-AR mRNA的表达,预先24 h给予普伐他汀(0.01,0.1,1 mmol·L^(-1))能剂量依赖性抑制醛固酮的上述作用,同时普伐他汀的这种抑制作用可被甲羟戊酸所逆转。结论:普伐他汀可抑制醛固酮诱导的心脏成纤维细胞ET-AR mRNA表达,其机制可能与甲羟戊酸代谢途径有关。 AIM: To investigate the effects of pravastatin on the expression of endothelin subtype A receptor (ET-AR) mRNA induced by aldosterone in cultured neonatal rat cardiac fibroblasts. METHODS: The expression of ET-AR mRNA was detected and quantified using reverse transcriptase-polymerase chain reaction (RT-PCR) method. RESULTS: Aldosterone 0.1 μmol·L^-1 significantly up-regulated ET-AR mRNA expression of cardiac fibroblasts, and the effect was blocked by pravastatin (0.01, 0.1, 1 mmol·L^-1) dose-dependently. In contrast, pravastatin-induced inhibitory effect was reversed by mevalonate. CONCLUSION: Pravastatin down-regulated ET-AR mRNA expression induced by aldosterone in a process specifically related to mevalonate in cardiac fibroblasts.
