摘要
目的:探讨姜黄素防治高脂血症致肾脏损害的作用机理。方法:喂食高脂饲料造成大鼠高脂血症致肾脏损害模型,姜黄素(高、中、低剂量)同时干预给药12周,实验结束后,分别检测大鼠肾皮质TGF-β1mRNA和MMP-2mRNA的表达。结果:姜黄素高、中剂量组肾皮质TGF-β1mRNA表达较模型对照组显著减少,且姜黄素高剂量组显著少于低剂量组(P<0.01);姜黄素高剂量组肾皮质MMP-2mRNA表达较模型对照组明显增多,且明显多于低剂量组(P<0.01)。结论:姜黄素能减少肾皮质TGF-β1mRNA表达,提高肾皮质MMP-2mRNA表达,呈量效依赖关系,从而有效地防治高脂血症致肾脏损害。
Objective: To explore the mechanism of cureumin in preventing and treating renal injury caused by hyperlipemia. Methods : Make the rats model of renal injury caused by hyperlipemia by feeding high - lipid forage, meanwhile, intervene by administering cureumin at high,medium and low dosage for 12 weeks. After the experiment, respectively test the expression of renal cortex TGF -β1 mRNA and MMP -2mRNA. Results: Compared with model control group ,the expression of renal cortex TGF -β1 mRNA in the high and medium dosage curcumin groups are decreased obviously( P 〈 0.01 ) ;the expression of renal cortex TGF -β1 mRNA in the high dosage curcumin group are much decreased, compared with low dosage cureumin group( P 〈 0.01 ) ;in the high dosage cureumin group,the expression of renal cortex MMP- 2mRNA is much more increased than that in model control group and low dosage cureumin group ( P 〈 0.01 ). Conclusion: The curcumin can inhibit the expression of renal cortex TGF -β1 mRNA and improve the expression of renal cortex MMP -2mRNA, in a dose -effect relationship, therefore can effectively prevent and treat the renal injury caused by hyperlipemia.
