芪藤通络饮对实验性糖尿病周围神经病变大鼠的防治机制

The Preventive and Therapeutic Mechanism of QiTeng TongLuo Decoction on Experimental Diabetic Neuropathy Rats

目的:探讨芪藤通络饮对糖尿病周围神经病变大鼠(diabetic perineuralpathies,DPN)的可能防治机制。方法:链脲佐菌素(STZ)诱导的糖尿病大鼠给予中药汤剂灌胃治疗,连续8周,观测体重、血糖、糖化血红蛋白的变化,观察药物对糖尿病大鼠坐骨神经形态学改变、用免疫组化的方法检测胰岛素样生长因子-1(IGF-1)在坐骨神经中的蛋白表达量。结果:中药组大鼠坐骨神经病变比模型组轻,轴突变性萎缩、髓鞘脱落较少,IGF-1在坐骨神经中的的表达与模型组相比显著性增高(P<0.05)。结论:本中药可改善糖尿病大鼠周围神经组织的代谢功能,提高神经元的存活,有利于神经的再生和修复,其作用可能是通过提高大鼠坐骨神经IGF-1蛋白的表达量来实现的。

Objective ： To study the preventive and therapeutic mechanism of QiTeng TongLuo decoction on rats with DPN （diabetic perineuralpathies ） induced by STZ. Methods ： Rats were fed with QiTeng TongLuo decoction through stomach eight weeks continuously. Changes on body weight, blood glucose,HbAlc were measured. Morphologic changes on sciatic nerves were also observed. Immunohistochemistry was used to detect the expression of IGF - 1 （ insulin - like growth factor - 1 ） in sciatic nerves. Results ： Pathological changes of sciatic nerves in Chinese herb group were obviously lighter than model group, denaturation and atrophia of neuraxon and ecclasis of medullary sheath was less; expression of IGF - 1 in sciatic nerve was significantly higher than model group. Conclusion ： QiTeng TongLuo decoction could improve the metabolism of perineural tissues of diabetic rats, enhance the survival and promote the regeneration and restoration of nerves, its function maybe achieved through increasing the expression of IGF - 1 in sciatic nerve of diabetic rats.
