摘要
目的:观察1(2,6二甲基苯氧基)2(3,4二甲氧基苯乙胺基)丙烷盐酸盐(DDPH)对实验性高血压大鼠血管平滑肌细胞(VSMC)增殖的作用及对生长因子PDGFB,bFGF及其相关癌基因csis,cmyc表达的影响。方法:采用氚胸腺嘧啶核苷(3HTdR)掺入法,电镜,免疫组化,原位杂交法进行实验观察。结果:DDPH在降低血压同时,能减少VSMC的线粒体,粗面内质网和3HTdR掺入量,并能逆转VSMC增殖时PDGFB,bFGF抗原及csis,cmycmRNA的表达增强。结论:DDPH能抑制实验性高血压大鼠的VSMC增殖,与生长因子及癌基因调控的分子生物学机制有关。
OBJECTIVE: To determine the effects of 1 (2, 6 dimethylphenoxy) 2 (3, 4 dimethoxyphenylethy lamino) propane hydrochloride (DDPH) on proliferation of vascular smooth muscle cells (VSMC) and the expression of growth factor PDGF B, bFGF and oncogene c sis, c myc in experimental hypertensive rats. METHODS: 3 H thymidine ( 3 H TdR) incorporation, electronic microscopy, immunohistochemistry and in situ hybridization assaies were used. RESULTS: After administration of DDPH, morphological change of VSMC was antagonized, and blood pressure and 3 H TdR data were significantly reduced. Furthermore, DDPH reversed the enhanced expression of antigen PDGF B, bFGF and reduced expression of oncogene c sis, c myc in experimental hypertensive rats. CONCLUSION: These results suggested that DDPH might inhibit proliferation of VSMC in experimental hypertensive rats, related to the mechanism of molecular biology of controlling growth factors and oncogene.
