摘要
目的:检测一种恶性疟原虫保护性抗原重组复合基因表达产物(HRA)诱导小鼠细胞免疫的反应.方法:免疫小鼠的脾淋巴细胞,体外分别经ConA、HRA和可溶性裂殖子抗原(SMA)的刺激,3H-TdR参入法检测淋巴细胞的增殖水平.结果:含佐剂的HRA免疫BALB/c和C57BL/6小鼠后,其脾细胞经HRA刺激后,增殖明显,可溶性裂殖子抗原免疫的BALB/c小鼠脾细胞经HRA刺激发生明显的增殖.结论:HRA上的T细胞位点发挥了效应.具有一定的突破MHC限制的能力.
Objective: The cellular immune responses to a Plasmodium falciparum hybrid recombinant antigen (HRA) were investigated. HRA codes for several T/B cell epitopes from MSA1, MSA2, RESA and Universal T cell epitopes of IL 1, tetanus toxiod. Methods: Splenocyte proliferation of immunized mice was examined by 3H TdR incorporation. Results: HRA could prominently induce splenocyte proliferation of BALB/c and C57BL/6 mice after immunized with HRA emulsified with Freund adjuvant.HRA could also induce prominently proliferation BALB/c mice immunized with soluble merozoite antigen (SMA). Conclusion: T cell epitopes of HRA are effective. HRA may overcome MHC restriction.
