摘要
目的研究奥美拉唑作为主动外排泵抑制剂提高铜绿假单胞菌对氧氟沙星的敏感性。方法二倍稀释法筛选临床分离的铜绿假单胞菌,测定耐氧氟沙星菌株对主动外排泵抑制剂碳酰氰基-对-氯苯腙(CCCP)和加入奥美拉唑后的敏感性。将CCCP和奥美拉唑分别加入铜绿假单胞菌培养液,分为单纯培养的铜绿假单胞菌组、铜绿假单胞菌+CCCP组、铜绿假单胞菌+奥美拉唑组,用RP-HPLC测定分别加入CCCP和奥美拉唑后的0.17、0.50、0.75、1、3、8、15、30、60 min时铜绿假单胞菌菌体内摄取氧氟沙星的含量。结果在不同时间点,铜绿假单胞菌+CCCP组和铜绿假单胞菌+奥美拉唑组菌体内摄取的药量均显著高于铜绿假单胞菌组。结论奥美拉唑作为主动外排泵抑制剂,可增强铜绿假单胞菌对氧氟沙星的敏感性。
OBJECTIVE To study the inhibition effect of Omeprazole on active efflux pump of Pseudomonas aeruginosa in Ofloxacin resistant strains. METHODS Resistant P. aeruginosa were screened from clinical isolates by using two- double dilution method. CCCP and Omeprazole were used as inhibitor of active efflux pump for detecting the resistant mechanism. The concentration of Ofloxacin ingested by P. aeruginosa at different times ( 0. 17,0. 5,0. 75,1,3,8,15,30 min) was measured by RP - HPLC. The P. aeruginosa groups were compared with P. aeruginosa + CCCP groups and P. aeruginosa + Omeprazole groups. RESULTS P. aeruginosa + CCCP groups and P. aeruginosa + Omeprazole groups were both higher than P. aeruginosa groups in all time points. CONCLUSION Omeprazole can inhibit the active efflux pump of P. aeruginosa in Ofloxacin resistant strains.
