摘要
目的建立测定人血清中头孢他啶浓度的方法。方法溶液为30mmol·L^-1硼砂(pH9.03)、电泳电压16kV、检测波长254nm,人血清样品经甲醇沉淀、离心、过滤后直接进样。结果头孢他啶浓度0.79~19.15mg·ml^-1与峰面积呈线性关系(r=0.9800),平均方法回收率为105.9%,RSD=6.45%(n=5),最低检测浓度为6.2μg·ml^-1,日内精密度RSD〈6.1%。结论所用方法具有简便、快速、准确的特点,可作为头孢他啶及其代谢过程中浓度的测定方法。
OBJECTIVE To establish a method for determination of Ceftazidime concentration in human serum. METHODS A new capillary zone electrophoresis method was adopted. 30 mmol·L^-1 sodium tetraborate ( pH9. 03 ) was used as experimental buffer. The voltage was 16 kV and the UV detection wavelength was 254 nm. The serum was precipitated with methanol and centrifuge before infection. RESULTS Calibration curve for ceftazidime was linear within the range of 0. 79 to 19. 15 mg·ml^-1 ( r = 0. 9800). The method recovery was 105.9% and intra - day RSD was less than 6. 1% (n = 6). CONCLUSION This method is simple, rapid and accurate. It can be successfully used for the determination of ceftazidime concentration in human serum.
