摘要
目的采用GV核酸染料取代溴化乙锭(EB)进行核酸染色,观察DNA染色结果,证实采用改进的核酸染色方法可替代EB进行常规核酸染色。方法提取小鼠肝组织DNA,采用GV与EB分别进行染色和电泳,在紫外凝胶成像仪下观察和记录结果。结果在紫外凝胶成像仪下,可观察到经两种方法染色的DNA条带清晰,亮度相当。结论GV是一种安全灵敏的核酸染料,可替代EB进行常规核酸染色。
Objective To stain nucleic acid by gold view(GV) for substituting ethidium bromicle(EB), to investigate the staining DNA and to prove that GV can substitute EB for routine nucleic acid staining. Methods DNA of rat liver tissue was extracted and stained with GV and EB. The staining results were invesatigated under UV transilluminator. Results Under the UV transilluminator, DNA strips stained by GV could be seen with the same brightness as those stained by EB. Conclusion Gold view is a safe, sensitive alternative to EB for routine nucleic acid staining.
