葛根素对脑缺血再灌注家兔血浆MDA、SOD含量的影响

摘要 目的观察葛根素对脑缺血再灌注损伤家兔血浆MDA、SOD含量的影响。方法健康家兔24只,随机分为假手术组(A组)、缺血再灌注组(B组)和葛根素处理组(P组),每组8只。利用"六血管"模型阻断动脉30 min,恢复血流再灌注4 h。P组于缺血前10 min静注葛根素5 mg/kg,继之以微泵150μg/(kg.min)输注至实验结束,B组不用药。观察缺血前15 min和再灌注4 h家兔血浆MDA及SOD浓度的变化。结果B组MDA较A组显著升高,SOD显著低于A组(P<0.01);与B组比较,P组SOD回升,而MDA显著降低(P<0.01)。结论葛根素能降低缺血再灌注期家兔血浆MDA的含量,提高SOD的活力,减轻脑缺血再灌注损伤。 Objective It is to observe the influence of puerarin on plasma malondialdehyde (MDA), superoxide dismutase (SOD) in rabbits following cerebral ischemia-reperfusion injury. Methods 24 rabbits were randomly divided into sham operation group(group A, n = 8) ,ischemia - reperfusion group (group B, n= 8) and puerarin group (group P, n = 8) with whole cerebral ischemia induced by clipping of bilateral external, internal carotid arteries and vertebral arteries for 30 min and then with the reperfusion lasting 4 hours. Group P with puerarin (Smg/kg) 10 min before occlusion and followed 150? g/(kg· min) intravenously till the end of procedure. The levels of plasma MDA and SOD were assayed 15 min before occlusion (TO) and 4 hours after reperfusion (T4). Results Compared with the group A, the levels of MDA were increased and the levels of SOD were decreased significantly in group B (P all 〈 0.01). Compared with the group B, the levels of MDA were increased, and the levels of SOD were decreased significantly in Group P (P all〈0.01 ). Conclusion Puerarin can attenuate the brain reperfusion injury, through suppresses the production of MDA, and improves SOD activity.
