rhGH在体外对人肝癌细胞系的作用及其Cyclin D_1蛋白表达的影响

摘要 目的研究重组人生长激素(rhGH)在体外对人肝癌细胞系SMMC7721生长的作用及其Cyclin D1蛋白表达的影响。方法实验分为对照组、rhGH组、rhGH+阿霉素(ADM)组及ADM组;通过体外细胞培养、MTT比色技术及免疫细胞化学技术等手段,测定不同浓度的rhGH及其与ADM合用时对人肝癌细胞系SMMC7721的生长曲线、细胞抑制率及其Cyclin D1蛋白表达的影响。结果与对照组相比,不同浓度的rhGH在体外对人肝癌细胞系SMMC7721的分裂增殖无明显影响,生长曲线、Cyclin D1蛋白表达不随rhGH浓度的增加而升高;rhGH+ADM组与ADM组比较差异无显著性,生长曲线、Cyclin D1蛋白表达无显著变化;与对照组及rhGH组相比较,单用ADM或rhGH+ADM对该细胞的抑制率增加,Cyclin D1蛋白表达降低(P<0.01),而两组间差异无统计学意义。结论rhGH在体外对人肝癌细胞系SMMC7721的分裂增殖无明显促进作用,对Cyclin D1蛋白表达无明显影响。 Objective To investigate the effects of recombinant human growth hormone (rhGH) on growth of human hepatic carcinoma cell line SMMC7721 and its expression of Cyclin D1 in vitro. Methods Four groups were assigned as control group, rhGH group, rhGH + ADM group and ADM group. By cell culture, MTF assay and inrnunocytochemistry, effects of different concentrations of rhGH and/or in colnbination with ADM on the cured line of cells growth, cell inhibitory rate and Cyclin D1 protein expression of human hepatic carcinoma cell line SMMC7721 were studied. Results In vitro the distinctly accelerated effects of different concentrations of rhGH on multiplication of SMMC7721 cell line were not found. It was not found that the cured line of cells growth and the expression of Cyclin D1 protein rose when compared with control group. There was no statistical difference on the cell proliferation between rhGH + ADM group and ADM group ( P 〉 0.05 ), and the cured lines of cells growth and the expression of Cyclin D1 protein have no difference. Compared with control group and rhGH group, cell inhibition rate remarkably increased and the expression of Cyclin D1 protein significantly decreased in rhGH + ADM group or ADM group ( P 〈 0.01 ), and the two groups have no difference in cell inhibition rate and the expression of Cyclin D1 protein ( P 〉 0.05 ). Conclusion In vitro rhGH does not distinctly accelerate the multiplication of human hepatic carcinoma cell line SMMC7721, furthermore it does not significantly affect the expression of Cyclin D1 protein.
