摘要
目的探讨奥曲肽联合透明质酸钠对兔术后腹膜粘连TGF-β1表达的影响。方法建立兔术后腹膜粘连模型,然后分成4组:对照组、透明质酸钠组、奥曲肽组、透明质酸钠联合奥曲肽组;术后14天剖腹手术,取粘连部位组织进行SP免疫组化染色,检测TGF-β1的表达。采用图像分析系统进行分析,测定TGF-β1的积分光密度(IOD)值,判定TGF-β1的表达量。结果奥曲肽组较对照组、透明质酸钠组TGF-β1的表达显著降低(P<0.001);联合治疗组亦较对照组、透明质酸钠组TGF-β1的表达显著降低(P<0.001)。奥曲肽和透明质酸钠在抑制TGF-β1的表达上无协同作用(q<1)。结论奥曲肽可通过抑制TGF-β1的表达预防腹膜粘连,而透明质酸钠具有隔离作用,两者作用于腹膜粘连发生过程中的不同环节发挥协同作用。
Objective To investigate the effect of combination of Octreotide and sodium hyaluronate on TGF - β1 in postoperative intraperitoneal adhesions of rabbits. Method Forty eight of Newzeland rabbits were used to establish the postoperative abdominal adhesion model and divided into four groups: model control group ; sodium hyaluraronate group; Octreotide group;octreotide and sodium hyaluronate group. Laparotomy operation after 14 days, the expression of TGF -β1 on the samples obtained from the adhesion sites were used by Immunohistoche mistric method and quantitative analysis of immunostaining. Results Octreotide group was lower than the model control group and the HA group on the expression of TGF - β1 on the samples obtained from the adhesion sites( P 〈 0. 001 ). Combination of octreotide and sodium hyaluronate have no synergetic effect on the inhibitory expression of TGF -β1 protein(q 〈 1). Conclusions The preventive action of octreotide on the abdominal adhesion is mostly involved in the inhibitory expression of TGF-β1, protein, and that of sodium hyaluronate is mostly isolation action both of which interfered with two progress step of the intraperitoneal adhesion iniliation and produced synergetic action.
