摘要
目的探讨热休克蛋白-70(HSP70)对急性白血病(AL)预后的影响及与多药耐药基因mdr-1的关系。方法应用反转录聚合酶链反应半定量分析50例初治AL患者(其中难治组10例,缓解组40例)细胞中HSP70、mdr-1的表达水平。结果难治组HSP70表达水平明显高于缓解组(P<0.01);mdr-1阴性组HSP70表达水平明显低于mdr-1阳性组;mdr-1 mRNA的表达与HSP70 mRNA的表达呈显著正相关。结论HSP70 mRNA的高表达是AL疗效差、预后不良的标志,且HSP70与mdr-1协同影响耐药。
[ Objective] To investigate the correlation between heat shock protein 70 (HSP70) and multidrug resistance-1 (mdr-1) and effect of HSP70 expression on prognosis in acute leukemia (AL). [ Methods] Expression level of HSP70 and mdr-1 was determined in 50 newly diagnosed AL patients by reverse tyanscription polymerase chain reaction (RT-PCR). [ Results] The expression of HSP70 in refractory AL was significantly higher than that in AL patients in complete remission (CR). The expression of HSP70 was significantly decreased in patients with mdr-1 negative group than that with mdr-1 positive group. The expression of mdr-1 was significantly positively correlated with the concentration of HSP70. [ Conclusion] High expression of HSP70 may be a poor prognostic factor to AL patients. HSP70 probably influence drug re- sistance coordinated with mdr-1.
