氧化苦参碱对喉癌细胞株Hep-2抑制增殖及诱导凋亡的实验研究 被引量:8

摘要 目的观察氧化苦参碱(Oxy)对喉癌细胞株(Hep-2)的抑制增殖及诱导凋亡作用。方法培养喉癌细胞株(Hep-2),MTT比色法及流式细胞术测定Oxy对喉癌细胞株Hep-2抑制增殖及诱导凋亡的作用。免疫沉淀法纯化蛋白并ERK1/2试剂盒测定细胞外调节信号激酶(ERK)活性;Western blot测定p-ERK、Bax及Bcl-2表达。结果Oxy明显抑制Hep-2细胞增殖,诱导Hep-2细胞凋亡;Oxy可抑制ERK活性及p-ERK表达,同时上调Bax表达,降低Bcl-2表达。结论Oxy可以抑制Hep-2增殖,机制与抑制ERK活性及p-ERK表达有关;同时可诱导凋亡,其途径与上调Bax表达,降低Bcl-2表达有关。 [ Objective] To observe the effects of Oxymatfine on the proliferation and apoptosis of Hep-2 cell line. [ Methods ] The effect of Oxymatrine on the proliferation of Hep-2 ceils was observed by MTT assay. Detection of apoptosis was performed with flow cytometry. ERK activity was measured by immuno-precipitation, the expressions of p-ERK, Bax and Bcl-2 were analyzed by Western blot. [ Results] Oxymatfine can depress the proliferation of Hep-2 cell line. The intra- cellular ERK activity and p-ERK expression were decreased by different concentrations of Oxymatrine. Moreover, Oxymatfine can induce apoptosis of Hep-2, the mechanisms were correlated with upregulation of Bax expression and downregulation of Bcl-2 expression. [ Conclusion] Oxymatfine can depress the proliferation of Hep-2 ceil line with inhibiting activity of ERK and expression of p-ERK. Meanwhile, Oxymatrine can induce apoptosis of Hep-2 ceil line by enhancing Bax expression and depressing Bcl-2 expression.
