紫外照射诱导采后番茄苯丙氨酸解氨酶的分离纯化 被引量:9

摘要 为了解苯丙氨酸解氨酶(PAL)在紫外诱导采后果蔬抗病性中的作用,以番茄为试材,经2.4kJ/m2短波紫外线(UV-C)照射,取果皮经冰浴研磨、硫酸铵分级沉淀、凝胶过滤和离子交换层析,分离纯化PAL,测得UV-C照射组PAL总蛋白含量为0.030mg,较对照组高0.002mg,酶比活为11466.0U/mL,比对照组高1109.0U/mL,分离纯化倍数为16.4倍,对照为15.0,表明UV-C照射可诱导PAL产生,提高活性。纯化获得的PAL经SDS-聚丙烯酰胺凝胶电泳分析,呈现一条带,其分子量约为304kD。 Phenylalanine anmlonia-lyase(PAL)of tomato peel were studied after 2.4 kJ/m^2 ultraviolet light-C (UV-C) irradiation in order to known the roles of PAL inducing disease resistance in post-harvest fruits and vegetables. PAL was purified by ice rubbing, sulfate grade deposition, gel filtration and ion exchange chromatography. Induced PAL total proein content was 0. 030 mg, more than 0. 002 nag to control group, PAL activity was 11 466. 0 U/mE, more than 1 109.0 U/mL to control group. Purification multiple was 16.4 and the control was 15.0. It showed PAL was induced to produce after UV-C irradiation. The enzyme was purified with SDS-PAGE and a single albumen strip was obtained, its molecular weight was identified to 304 kD.
