摘要
目的通过壳聚糖酶固定化前后最适宜的反应温度、pH值、米氏常数等动力学性质的变化研究载体电荷极性、空间阻力对壳聚糖酶动力学性质的影响。方法以两种极性不同的物质(几丁质、DEAE纤维素)为载体、戊二醛为交联剂,对本实验室自制的壳聚糖酶进行了固定化,然后分别分析酶经固定化后最适宜的反应温度、pH值、米氏常数等动力学性质的变化。结论酶经固定化后所产生的空间扩散阻力对壳聚糖酶的动力学性质的影响要比在固定化过程中所用载体所带的电荷的极性对酶的动力学性质的影响大的多。
Objective: To explore the effect of interspace diffuse resistance and carrier's electrical property on immobi- lized ehitosanase's kinetic property. Methods: ehitosanase was immobilized on two earlier which have different electrical property with glutaraldehyde by crossing - linking reaction. The change of the optimal temperature and pH , Kinetic param- eter of the immobilized enzyme were studied. Conclusion: The effect of interspace diffuse resistance for chitosanase's kinetic property is found larger than that of carrier's electrical property.
