摘要
目的:探讨卵巢癌细胞系COC1、COC1/DDP、A2780、A2780/DDP中切除修复交叉互补基因(ERCC1)表达与顺铂耐药的关系。方法:采用RT-PCR方法检测4种卵巢癌细胞系中ERCC1基因的表达,MTT检测各细胞系对顺铂的敏感性。结果:4种细胞系中均有ERCC1基因表达,耐药细胞中的表达量明显高于敏感细胞(P<0.05)。DDP作用于COC1、COC1/DDP细胞72小时后,随着ERCC1基因表达的升高,细胞对顺铂的耐药性增加。结论:卵巢上皮性癌细胞系中ERCC1基因表达的升高与卵巢癌顺铂耐药相关。
Objective:To study the relationships between ERCC1 gene expression and Cisplatin resistance in COC1 ,COC1/DDP,A2780 and A2780/DDP ovarian cancer cell lines.Methods:RT-PCR was used to detect the expression of ERCCI mRNA in ovarian cancer cell lines and MTT assay to detect the sensitivity of each cell line to Cisplatin.Results:The 4 cell lines all had the expression of ERCC1 mRNA ,and the expression of ERCC1 mRNA in drug resistance cell lines was significantly higher than that of the sensitive cell lines (P〈0.05).After being effected by DDP for 72h,MTT assay showed that the Cisplatin resistance increased with the increasing of ERCC1 expression.Conclusions:The expression of ERCC1 mRNA in ovarian cancer cell lines correlates with Cisplatin resistance.
