摘要
目的探讨培哚普利对肾性高血压大鼠心肌纤维化作用及对转化生长因子-β1表达影响。方法建立双肾一夹肾血管性高血压(2K1C)大鼠模型,将24只实验大鼠随机分为假手术组、高血压组、培哚普利组,术后4周末开始灌胃培哚普利片2mg/(kg.d),共8周。测量大鼠尾动脉收缩压、心脏重量/体重、心肌胶原容积分数(CVF)、血管周围胶原容积分数(PVCA)及转化生长因子-β1(TGF-β1)的表达。结果和假手术组相比,高血压组的收缩压、心脏重量/体重、CVF及PVCA显著增加(P<0.01),TGF-β1表达上调(P<0.01)。和高血压组相比,培哚普利组心脏重量/体重、CVF、PVCA及TGF-β1的表达降低(P<0.05)。结论培哚普利可以减轻肾性高血压大鼠心肌纤维化,其机制可能与下调TGF-β1有关。
Obiective To investigate the effects of Perindopril on myocardial fibrosis and expression of transforming growth factor - β1 (TGF- β1 ) in renovascular hypertenslve(RH) rats. Methods RH models were established in 24 rats, rats were randomly divided into sham group,RH group and Perindopril group. After four weeks, rats in perindopril group were administered perindopril 2 mg/(kg· d) for eight weeks. Systolic blood pressure (SBP)was measured, cardiac mass index,collagen volume fraction(CVF) ,perivascular collagen area(PVCA) and the expression of TGF-β1 in left ventricular tissue were calculated. Results Compared with sham group, The SBP, cardiac mass index, CVF, PVCA and expression of TGF-β1 increased markedly in RH group(P〈0.01). Compared with the RH group, cardiac mass index , CVF, PVCA and expression of TGF-β1 decreased in perindopril group(P 〈 0.05). Conclusion Perindopril could reverse myocardial fibrosis in renovascular hypertensive rats. It was probably related to decrease expression of TGF- β1.
