摘要
目的:运用流式细胞术检测苦参素对人胃癌细胞系MGC-803凋亡的作用。方法:运用流式细胞术检测细胞凋亡率、凋亡峰及凋亡相关基因蛋白Bcl-2和Bax的表达水平。结果:苦参素1mg/mL、2mg/mL作用24h后,流式细胞仪(FCM)检测结果显示,细胞凋亡率与阴性对照组相比,差异具有显著性(P<0.05);Bcl-2表达量明显减少,Bax表达量明显增加,差异具有显著性(P<0.05)。结论:苦参素对人胃癌细胞系MGC-803有诱导凋亡作用,其机制与上调Bax和下调Bcl-2表达有关。
Objective:To detect the action of kurarinone on the induction of apoptosis of human gastric carcinoma cell line MGC-803 by flow cytometry. Methods:The apoptotic rate and peak as well as the expression level of apoptosis-related gene Bcl-2 and Bax were detected by flow cytometry. Results: Compared with negative controls, pronounced difference (P〈0.05) was detected by flow cytometry in apoptotic rate after the use of 1 mg/mL, 2 mg/mL kurarinone for 24 hours. A marked drop in Bcl-2 level and a marked increase in Bax level were observed, accounting for a significant difference (P〈0.05). Conclusion: Kurarinone induces apoptosis of human gastric carcinoma cell line MGC-803. Its mechanism is asso- ciated with up-regulating of Bax gene and down-regulating of Bcl-2 gene.
