柞蚕雄蛾提取液改善W256荷瘤大鼠放疗后免疫功能的实验研究 被引量:2

摘要 目的观察柞蚕雄蛾提取液对荷W256癌肉瘤大鼠放疗后免疫功能的影响。方法60只Wistar大鼠随机分为正常对照组、模型组、单纯放疗组和实验组(放疗与柞蚕蛾提取液联合治疗组),每组15只。建立W256荷瘤大鼠模型,分离脾脏和胸腺,并计算脏器指数;采用流式细胞术(FCM)及酶联免疫吸附试验(ELISA)分别检测外周血T淋巴细胞亚群变化和血清中细胞因子INF-γ和IL-4含量。结果与放疗组相比较,柞蚕雄蛾提取液可以升高放疗后荷瘤大鼠脾脏和胸腺指数,增加放疗后荷瘤大鼠外周血中CD4+T细胞比例与CD4+T/CD8+T值(P<0.05);实验组荷瘤大鼠血清中细胞因子INF-γ含量明显升高,而IL-4含量显著降低(P<0.05)。结论柞蚕雄蛾提取液可以逆转荷瘤大鼠放疗后免疫功能抑制状态,改变荷瘤机体免疫功能,可作为恶性肿瘤传统放疗中的辅助用药。 Objective To study the effects of an extract from the male zooid on reversing the radiation-induced im- munosuppression of rats bearing W256 carcinoma. Methods Tumor models were established by subcutaneously injecting W256 tumor cells into rots. 60 Wistar rots were randomly divided into 4 groups. The thymus index and the spleen index were used as parameters to evaluate the effect of an extract from the male zooid on non-specific and specific immune function of rats with tumor. The presence of Interferongamma (INF-γ) and interleukin-4 (IL-4) in the peripheral blood serum were screened by enzyme-linked immunosorbent assay (ELISA) to investigate cell-mediated immune responses and T subsets were determined by flow cytometry(FCM). Results The extract from the male zooid significantly raised the thymus index and the spleen index of rots with tumors. The number of CD4^+ T and the CD4^+ T/CD8^+ T ratio of W256-bearing rots treated with the extract from the male zooid significantly increased compared with the radiotherapy group ( P 〈 0.05). Also, the level of Interferon-gamma (INF-γ) was up-regulated and interleukin -4(IL-4) was down-regulated. Conclusions The extract from the male zooid of Antheraea pemyi has effect of reversing the radiation-induced and couldbe regarded as a main assistant agent combined with traditional radiotherapy for treatment of malignant tumors.
