重组小鼠γ-干扰素对弓形虫感染孕鼠及鼠胚的影响 被引量:2

摘要 目的研究重组小鼠γ-干扰素(rmu~IFN-γ)对妊娠期感染弓形虫的孕鼠蜕膜T淋巴细胞亚群及鼠胚感染率的影响。方法将60只孕鼠被随机分为4组:空白组、感染组、rmu—IFN-γ小剂量治疗组(A组)和大剂量治疗组(B组)。采用弓形虫急性感染妊娠第6~10d孕鼠,不同剂量rmu—IFN-γ腹腔给药,感染后72h取孕鼠蜕膜组织,流式细胞术检测各组T淋巴细胞亚群的水平;每组随机取5只孕鼠统计各组鼠胚的数目,SABC免疫组化法检查鼠胚中的弓形虫抗原阳性率。结果孕鼠感染弓形虫后蜕膜CD4^+、CD4^+/CD8^+降低(P〈0、05),A组和B组两种剂量的rmu—IFN-γ干预后均能提高CD4^+、CD4^+/CD8^+水平(P〈0、05);A组和B组中鼠胚的垂直传播率均低于感染组。结论孕鼠感染弓形虫后蜕膜局部细胞免疫受到抑制,注射适量的rmLl—IFN-γ可以逆转CD4^+/CD8^+比值,提高机体的细胞免疫水平,降低鼠胚的垂直传播率,对孕鼠及鼠胚具有-定的保护作用。 Objective To explore the effects of rmu - IFN - γ-on T ceil subsets in decidual tissues of pregnant mice infected with toxoplasma gondii. Methods Pregnant mice, infected with T. gondii, were injected intraperltoneally rmu - IFN - γand then killed after infected 72 hours , The numbers of CD4^ + and CD8^+ cell in peripheral blood and decidual tissues were detected by flow cytometry; The incidence of maternal -fetus transmission was calculated by immunohistochemical staining of toxoplasma gondii antigen, Results The level of CD4^+ cells,CD4^+/CD8^+ both in decidual tissues of the group infected with T. gondii was lower than that of control group during the gestation ( P 〈 0. 05 ). The level of CD4^+ cells, CD4^+/CD8^+ in mice administered with different dosages of rmu - IFN - γ rose up dramatically( P 〈 0.05 ), the rate of maternal - fetus transmission of both two administered groups was lower than infected group. Conclusion The peripheral cellular immunity of pregnant mice was suppressed after infected with T, gondii. A proper dose of rmu - IFN - γ could have reversed the decline of the ration of T cell subsets, thus elevated the level of peripheral cellular immunity of pregnant mice. also reduced the rate of maternal - fetus transmission of toxoplasma gondii, so it may play an important role in protection of pregnant mice and embryos.
