全反式维甲酸对Tenon囊成纤维细胞超微结构的影响

摘要 目的观察全反式维甲酸(ATRA)对体外培养的人Tenon囊成纤维细胞(HTCFs)超微结构的影响。方法取第3~6代HTCFs细胞,用1×10-5mol/L的ATRA处理细胞3d后,于扫描电镜下观察细胞表面结构,透射电镜下观察细胞超微结构的变化。结果扫描电镜下经ATRA作用后人Tenon囊成纤维细胞趋于变圆,表面微绒毛几乎消失,可见起泡现象;透射电镜下细胞微绒毛消失,胞内大量空泡形成,胞质浓缩,核仁消失,核染色质固缩、边集,呈境界分明的新月形、块形,聚集于核膜下,线粒体肿胀、内质网扩张呈空泡状,但胞膜、核膜及各细胞器膜均保持完整,呈典型的凋亡特征。结论ATRA可明显影响HTCFs的超微结构,并出现细胞凋亡的特征性改变。 Objective Retinoic acid,a naturally occurring oxidative product of vitamin A,is known to have an effect on differentiation and proliferation of many types of cells. Our previous study has shown that all trans-retinoic acid (ATRA) can inhibit the growth of cultured human Tenon' s capsule fibroblasts(HTCFs). This study aimed to evaluate the effect of ATRA on the ultrastructure of HTCFs in vitro. Methods The Tenon' s capsule tissue from fresh corpse eye was obtained and cultured. The third to sixth passage of HTCFs were treated with ATRA (1 x 10-s tool/L)for 3 days, and control group was treated with anhydrous ethanol( 〈0.01% )for 3 days. Cell specimens were prepared and the superficial structure and uhrastructure of cells were observed under the scanning and transmission electron microscope. Results Under the electron microscope, the shape of HTCFs in control group was shuttle-like with microvilli on the surface, and the cell organs were intact and the nucleoli were clear. Typical apoptotic cellular changes were found after ATRA treatment, showing the rounding of cells, disappearance of the microvilli on the cell surface,the vacuolar change of cellular membrane, concentration and margining of chromatin, edema and vacuolation of mitochondria and endoplasmic reticulum. However,the cell membrane, nucleus and cellular organs were intact. Conclusion ATRA can induce apoptosis of human Tenon' s capsule fibroblasts.
