肌肉素干预大鼠骨骼肌葡萄糖摄取与NF-κB的相关性 被引量:7

The Relevance Between Muscle Intervention for Rat Skeletal Muscle Glucose Uptake and the Expression of NF-κB

摘要 采用同位素示踪法检测3H标记-葡萄糖(3H-G)摄取,半定量RT-PCR检测NF-κBI、L-6 mRNA的表达以研究肌肉素(musclin)干预大鼠骨骼肌葡萄糖摄取的机制。结果显示,musclin降低了胰岛素诱导大鼠分化的成肌细胞葡萄糖摄取,NF-κBI、L-6 mRNA的表达水平升高。肌肉素降低胰岛素诱导大鼠骨骼肌葡萄糖摄取,可能与其对NF-κB的激活作用有关。 Musclin impairs insulin-stimulated glucose uptake in the differentiation of C2C12 myocytes. In these cells, musclin reduce ^3H-G uptake, but the expression level of GLUT4 is not altered. Studies of the myocytes revealed that the expression level of NF-κB and IL-6 is increased, compared with that of myocytes preincubated in a musclin-free medium. The results indicate that musclin-induced insulin resistance can be studied in rat skeletal muscle in vitro. They suggest that musclin-induced impairment of insulin action in these muscles is related to NF-κB pathway.
