舒脉胶囊对实验性家兔肢体动脉硬化的防治作用及其机制的研究 被引量:3

摘要 目的观察舒脉胶囊对实验性家兔动脉硬化及脂代谢的影响,探讨该药防治肢体动脉硬化性闭塞症(ASO)的作用及机制。方法将家兔随机分为3组:正常组、模型组、给药组。用体内血栓形成测定仪对模型组和给药组家兔行右下肢髂动脉定位电流刺激术,术后第2天起饲喂高脂饲料(含3%猪油、1%胆固醇),术后第2天和第15天,分别对模型组和给药组注射10%的牛血清白蛋白250 mg/kg,以此复制ASO动物模型;正常组行假手术,饲喂普通饲料;术后第2天起给药组用舒脉胶囊水煎剂灌胃,正常组灌胃同体积的纯净水,实验周期60 d。经耳中动脉采血,检测血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)及高密度脂蛋白胆固醇(HDL-C)。同时观察髂动脉和胸主动脉的形态变化。结果①光镜:模型组血管内膜增厚、内膜下有大量的泡沫细胞堆积,管腔明显狭窄;而给药组可抑制内膜下泡沫细胞的生成,减轻内膜的增厚。②正常组胸主动脉内膜表面光滑,模型组血管内膜有大小不等的粥样斑块形成,管壁表面粗糙不光滑;给药组血管内膜局部有粥样斑块,但比模型组明显减轻。③模型组的血清CHO、TG、LDL-C、HDL-C显著升高,与正常组比较有显著性意义(P<0.01);而给药组能降低TG、LDL-C的含量,与模型组比较有显著性差异(P<0.05)。结论舒脉胶囊可缓解AS-ASO的形成,其作用机制与调控脂代谢有关。 Objective To investigate the effect of Shumai Capsules (SMC) on the earlier period atherosclerosis obliterans(ASO) in rabbits and to explore the mechanism of it. Methods Eighteen male Japanese albino rabbits were randomly divided into three groups, the control group, the model group and the SMC treatment group. The fight iliac arteries of the later two groups were stimulated by electric current generated by a thrombosis inducer, while the ones in the control group were treated with sham operation. From the day after the operation, the model group and the SMC treatment group were fed with high-fat diet(with 3% lard and 1% cholesterol) , the SMC group was given SMC by intragastric administration, and the control group was fed with the standardized rabbit food for all the period. On the 2nd and 15th day after the operation, the model group and the SMC treatment group were respectively injected with 10% bovine serum albumin 250 mg/kg. The whole experiment lasted for 60 days. When the experiment was over, the levels of serum total cholesterol (TC), triglyeride (TG), high density lipoprotein-cholesterol(HDL-C) and low density were harvested for the HE staining to thoracic aortae was stained with lipoprotein-cholesterol ( LDL-C ) observe the patho-appearance of magdala were detected. The iliac arteries the blood vessels and the wall of red Ill. Results ①Under the light microscope the iliac arteries in the control group showed integrated and continuation, and endothelial cells were lined up in order. The intima was thin. The model group showed a stegnosed lumens and a obviously thick intima filled with lots of foam cells full of fat. Compared with the model group, the theca interna from the SMC group was thinner, and the number of foam cell was fewer. ②In the control group, thoracic aorta intima was smooth, and in the model group thoracic aorta intima was rough with atheromatous plaques. In the SMC treatment group, atheromatous plaques were decreased significantly when compared with those in the model group. ③Compared with the control group, the levels of serum TC, TG, HDL-C and LDL-C were significantly increased in the model group(P 〈 0.01 ) ,while those results turned out a lower tendency in the SMC treatment group than in the model group. Significant differences only existed in LDL-C ( P 〈 0. 05)and TG(P 〈0.01 ). Conclusion SMC can relieve ASO by way of regulating lipid metabolism.
