摘要
目的观察艾麻口服液对慢性支气管炎大鼠气道炎症的影响,并探讨其部分疗效机制。方法用气管内注入低剂量脂多糖复制大鼠慢性支气管炎模型。雄性Wistar大鼠随机分为假手术组、慢支模型组、艾麻口服液组及橘红止咳口服液对照组(n=7)。测定各组大鼠支气管肺泡灌洗液(BALF)细胞总数及白细胞分类计数,肺组织中髓过氧化物酶(MPO)活力,BALF、血浆及肺组织匀浆液中总超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量,测定大鼠BALF、肺组织及外周血中肿瘤坏死因子-α(TNF-α)、白细胞介素-8(IL-8)的含量。结果慢性支气管炎大鼠BALF中炎性细胞明显增高,肺组织中的MPO活性、肺组织及外周血中MDA含量明显增加,BALF、肺组织及外周血中总SOD活力则明显降低;经艾麻口服液治疗后,BALF中炎症细胞分类计数、肺组织中MPO活性、肺组织及外周血中MDA含量均明显降低,BALF、肺组织及外周血中总SOD活力明显升高。进一步研究显示,艾麻口服液能减少慢支大鼠BALF中IL-8的含量,减少慢支大鼠BALF、肺组织匀浆液及血浆中TNF-α含量。结论艾麻口服液通过调节TNF-α、IL-8的分泌以减少慢支气道炎性细胞的浸润,同时具有促进氧自由基的清除和抗脂质过氧化损伤作用。
Objective To study the effects of Aima oral liquid ( AM ) on the airway inflammation in rats with chronic bronchitis(CB) and to study the therapeutic mechanism of AM. Method Rat model with CB was established by intratracheal injection with low dose of lipopolysaccharide. Male Wistar rats were randomly divided into four groups, sham operated(Sham) group, CB group, CB +AM group, and CB + Juhongzhike oral liquid (JH) group, with 7rats in every group. The total number of WBC and differential count in the bronchoalveolar lavage fluid (BALF) were counted. The activity of MPO in the lung, the activity of total SOD in BALF, lung and blood plasma, the MDA content in the lung and blood plasma, and the content of IL-8 and TNF-α in BALF, lung and blood plasma were measured. Results The number of total cells, monocyte-macrophage, neutrophil and lymphocyte were significantly increased in the BALF of model group compared with sham group (P 〈 O. O1 ). After treated by AM, the number of total cell and the and differential count WBC in the BALF were significantly decreased compared with CBrats (P 〈 0.05 ). The activity of total SOD decreased significantly in BALF, lung and blood plasma, and the content of MDA was increased significantly in BALF and lung in the CB rats ( P 〈 0.01 ) ; The activity was increased significantly and the content of MDA in CB + AM group (P 〈 0.05). Farther study showed that AM could significantly decreased the content of IL-8 in the BALF compared with CB model group, significantly decreased the content of TNF-ctcompared with CB model group, not only in the BALF, but also in the lung and blood plasma. Conclusion AM can reduce the airway inflammation of the CB rats through down regulating the secreting of TNFct and IL-8, and promoting the clearance of the oxygen free radical and increasing the ability to resist the lipoperoxidation injury.
