子宫内膜异位症患者在位内膜细胞的异常生物学特性 被引量:12

摘要 目的:探讨子宫内膜异位症(EM)患者在位子宫内膜细胞在侵袭、新生血管形成、增殖、凋亡诸多方面是否有别于正常内膜细胞。方法:1.应用ELISA方法对离体培养的人EM在位内膜细胞及正常子宫内膜细胞的培养基中血管内皮生长因子(VEGF)、基质金属蛋白酶-3(MMP-3)、基质金属蛋白酶组织抑制剂-1(TIMP-1)和细胞间黏附分子-1(ICAM-1)的浓度进行测定。2.应用逆转录聚合酶链反应对离体培养的人EM在位内膜细胞及正常子宫内膜细胞VEGF、MMP-9、TIMP-1和Bax的mRNA表达进行观察。3.应用流式细胞技术对离体培养的人EM在位内膜细胞及正常子宫内膜细胞的增殖和凋亡状况进行观察。结果:EM体外培养的在位内膜细胞分泌的VEGF和MMP-3均显著高于对照组(P<0.01,P<0.05);TIMP-1显著低于对照组(P<0.01);VEGF mRNA、MMP-9 mRNA表达显著高于对照组(P<0.01,P<0.05);TIMP-1 mRNA和Bax mRNA的表达均低于对照组(均P<0.05);增殖指数显著高于对照组(P<0.05);凋亡指数显著低于对照组(P<0.05)。结论:EM在位内膜细胞在表达分泌VEGF、MMP-3、MMP-9、TIMP-1、Bax,PI、ApI等诸多方面均有别于正常对照内膜细胞,进一步支持了"在位内膜决定论"。 Objective:To determine the abnormal biological characteristics of the cultured eutopic endometrial cells of endometriosis(EM).Methods:Vascular endothelial growthfactor(VEGF), matrix metalloproteinase-3 (MMP-3),tissue inhibitor of metalloproteinase-1 (TIMP-1),soluble intercellular adhesion molecule-1 (sICAM-1),VEGF mRNA,MMP-9 mRNA,TIMP-1 mRNA,Bax mRNA,proliferation index (PI) and apoptosis index (ApI) in the culture media of cells and in the cells themselves from EM patients and normal women were detected by ELISA,reverse transcriptation polymerase chain reaction (RT-PCR) and flow cytometry.Results:The levels of VEGF and MMP-3 in the culture media of cells from endometriosis were higher than that of control (P〈0.01, P〈0.05),however,the level of TIMP-1 of EM was less than that of control (P〈0.01).The expression of VEGF mRNA and MMP-9 mRNA were greater than that of control (P〈0.01,P〈0.05).While,the expression of TIMP-1 mRNA and Bax mRNA were less than that of control (both P〈0.05).The PI of cells from endometriosis was bigger than the control (P〈0.05),meantime,the ApI of endometriosis cells was lower than that of control (P〈 0.05).Conclusion:The potential of proliferate,secreting VEGF and MMPs of cultured EM eutopic endometrial cells is increased,comparing with that of normal endometrial cells. The apoptosis,on the contrary,is decreased.
