线栓法制备SD大鼠局灶性脑缺血/再灌注模型的改良研究 被引量:12

摘要 目的本研究的目的旨在对传统线栓法制备大鼠局灶性脑缺血/再灌注模型方法进行改良以提高实验成功率。方法选择SD大鼠50只,对传统制备方法进行改良。改良方法包括:1)激光多普勒流量仪监测脑血流;2)将脑血流严格降低到正常15%以下;3)对侧颈总动脉夹闭;4)选择头端5 mm包被硅橡胶直径0.35 mm的尼龙线作为栓线。结果脑血流监测可准确判断插线是否成功,当栓线插到合适位置时,脑血流迅速下降至30%以下,但很少降到15%以下;对侧颈总动脉夹闭,一般可将脑血流降至正常15%以下。造模成功率88%。结论改良后方法优于传统方法,制备模型过程中,严密监测脑血流、夹闭左颈总动脉以严格控制脑血流量至正常15%以下,是提高造模成功率的关键。应用头端包被硅橡胶的丝线对血管损伤小。 Objective The suture methods for the establishment of rat model of focal cerebral ischemia/reperfusion is used widely,but problems such as model unstability,low achievement ratio always exist.The Objective of this study is to improve the methods for getting successful of middle cerebral artery occlusion(MCAO) model.Methods Some newer methods were employed to overcome some drawbacks in traditional methods: 1) continuous laser doppler fluid meter was employed to monitor local cerebral blood flow,2) which was reduced to 15% of baseline, 3) the opposite common carotid artery was clamped,4) silicone rubber coated monofilament was selected to reduce the risk of vessel injury and intracranial bleeding.Results As soon as the filament got into the suitable position,the local cerebral blood flow would decreased sharply to 30% or less of baseline,but rarely to 15% of baseline.When the opposite common carotid artery was clamped simultaneously,the local cerebral blood flow would reduced to 15% or less of baseline.Monofilament of 0.35 mm diameter,5 mm coated with silicone rubber coated was more reliable than those we once used,because it had smooth,soft,and flexible tip,which could reduce the risk of vessel injury and intracranial bleeding.The model was produced success in 44 of 50 rats,achievement ratio was 88%.Conclusion The cerebral blood flow reduction to 15% or less of baseline is a key factor in suture methods for rat model of focal cerebral ischemia/reperfusion.Silicone-rubber coated monofilament is more reliable in the experiment.
