摘要
对海洋中筛选的酒香酵母(Brettanomycessp.)7-3产DHA条件进行了优化,结果表明培养基最佳组成为葡萄糖10 g/L,蔗糖10 g/L,酵母膏20 g/L,(NH4)2SO410 g/L,NaHCO31 g/L,K2HPO46 g/L,ZnSO40.1 g/L,MgSO40.1 g/L,CaCl20.5 g/L,Na2EDTA 0.2 g/L,FeC6H5O7.3H2O 0.01 g/L。摇瓶培养最佳条件为培养初始pH=6.0,培养温度25℃,装液量为100 mL(在500 mL摇瓶中),培养时间为84 h,优化后油脂与DHA产量分别提高了3.3和3.8倍。
The culture conditions of Brettanornyces sp. strain 7-3 screened from seawater wereop- timized for the production of DHA. The optimum medium (per liter) consisted of 10 g glucose, 10 g sucrose, 20 g yeast extract, 10 g (NH4)2SO4, 1 g NaHCO3, 6 g K2HPO4, 0.1 g ZnSO4, 0.1 g MgSO4, 0.5 g CaC12, 0.2 g Na2EDTA , and 0.01 g FeC6H5O7 · 3H20. The optimum fermenta- tion conditions were determined as follows, initial pH 6. 0, temperature 25 ℃, broth volume 100mL (i. e. , 20% for a 500-mL shake flask), and the culture time was 84 h, the productions of lipids and DHA were respectively increased by 3.3 times and 3.8 times.
