音乐对大鼠杏仁核神经元NMDA受体表达的作用

摘要 目的研究音乐刺激对大鼠杏仁核神经元NMDA受体表达的影响。方法在大鼠生长发育的不同时期,给予音乐刺激,用免疫组织化学、图像分析方法和体视学方法定量研究音乐刺激后,大鼠杏仁核神经元NMDA受体表达和NMDA受体免疫阳性细胞的密度变化。结果音乐刺激后,大鼠杏仁核神经元NMDA受体免疫染色的光密度比对照组增加,从对照组的(8.72±1.31)增加到持续音乐组(16.91±2.07),生后音乐组(15.52±1.71),生前音乐组(9.11±1.51);NMDA受体免疫阳性细胞的体积分数(%)、表面积密度(1/mm^2)和突起长度密度(1/mm^2),从对照组(5.64±1.23)%、(0.776±0.035)1/mm^2、(0.936±0.135)1/mm增加到持续音乐组(12.51±3.02)%、(1.513±0.353)1/mm^2、(2.527±0.436)1/mm,生后音乐组(11.64±3.23)%、(1.476±0.337)1/mm^2、(2.472±0.401)1/mm,生前音乐组(6.34±1.27)%、(0.796±0.041)1/mm^2、(1.077±0.142)1/mm,这些变化以持续音乐刺激组最明显,生后音乐刺激组次之,生前音乐刺激组和对照组最弱。结论音乐刺激能增强大鼠杏仁核神经元NMDA受体表达和NMDA受体免疫阳性细胞的密度,其增强作用具有刺激时间依赖性。 Objective To study the effect of music on expression of NMDA receptor on neuron from rat amygadale. Methods In different developing stages,the rats were exposed to music and the expression of NMDA receptor of neuron from the amygadale was tested with immunohistochemistry and image analysis, and the volume fraction, surface density and length density of neurons with positive NMDA receptor immunoreaction were examined quantitively with stereology. Results After music exposure,the optic density of NMDA receptor immunostaining on rat amygadale increased from 8.72±1.31 in control to 16.91± 2.07,15.52±1.71,9.11±1.51 in persist, postnatal and prenatal music exposure groups respectively; and the volume fraction( % ) ,surface density ( 1/mm^2 ) and length density ( 1/mm^2 ) of NMDA receptor immunostaining positive neuron from amygadale increased from 5.64±1.23,0.776±0.035,0.936±0. 135 in control,to 12.51±3.02,1.513±0. 353,2.527±0.436 in persist music exposure group, 11.64±3.23,1. 476±0.337,2.472±0. 401 in postnatal, and 6.34±1.27,0.796±0.041,1.077±0.142 in prenatal group respectively. All these parameters changed most in the persist music exposure group, then the postnatal, and least in the prenatal and control groups. Conclusion Music can enhance the expression of NMDA receptor and increase the density of NMDA receptor immunostaining positive neuron in amyg-adale with stimulating time-dependent manner.
