川芎嗪对急性心肌梗死模型大鼠的保护作用及机制

The protective effect and mechanism of ligustrate injection on acute myocardial infarct in rats

目的探讨川芎嗪对急性心肌梗死模型大鼠的保护作用及机制。方法利用结扎清醒大鼠冠状动脉左前降支复制急性心肌梗死模型，将大鼠随机分为假手术组，模型组、银杏叶提取物（20mg／kg）组及川芎嗪（10、20、40mg／kg）组。测定清醒大鼠心肌酶学、心肌梗死面积、细胞凋亡和组织病理HE染色的变化。结果急性心肌梗死模型大鼠SOD显著降低，MDA、LDH和心肌梗死面积显著增大，心肌细胞凋亡数量亦明显增多（P〈0.01～0．001）。川芎嗪能明显提高SOD、降低MDA、LDH的含量，减小心肌梗死面积和细胞凋亡数量（P〈0．05—0．001）。结论川芎嗪对急性心肌梗死模型大鼠具有一定的保护作用，其机制可能是通过缩小心肌梗死面积、改善心肌酶学、保护受损心肌细胞凋亡等多种途径实现的。

Objective Aim to study the protective effect and mechanism of ligustrate injection on acute myocardial infarct in rats. Methods The model of acute myocardial infarct was established by ligation of left anterior descending artery of the waking open chest rats. After the model was established, the rats were randonmy divided into 6 groups, they were sham operation group, model group, positive control group（ EGb 20 mg/kg）, LIg high-dose group（40 mg/kg）, LIg medial-dose group（20 mg/kg）and LIg low-dose group（ 10 mg/kg）. Measured myocardial e nzyInology, the area of myocardial infarct, apoptosis and the difference of HE stain. Results To the rats of model group, the activity of SOD were decreased , LDH, MDA and the area of myocardial infarct were reduced remarkably, the quantity of apoptosis were increased（ P 〈 0.01 - 0.001）.After treated by LIg （10 - 40 mg/kg ）, the activity of SOD were increased, LDH and MDA were reduced remarkably, the area of myocardial infarct and the quantity of apoptosis were decreased respectively（ P 〈 0. 05 - 0. 001 ）. Conclusion The results we observed suggest that LIg had protective effect on acute myocardial infarct in rats.The protective effect may be related to LIg＇s function of reducing the area of myocardial infarct, improving myocardial enzymology, decreasing apoptosis.
