癌基因C-K-ras和抑癌基因Rb突变与胰腺癌发生的关系

摘要 目的:探讨癌基因C-K-ras和抑癌基因Rb点突变与胰腺癌发生的关系。方法:选择经病理学证实的原发性胰腺癌19例、胰腺癌恶性淋巴瘤1例、胰腺多形细胞癌2例和慢性胰腺炎5例的石蜡包埋组织样本,采用PCR-SSP法检测C-K-ras 12位密码子和Rb第21位外显子的点突变情况。结果:19例原发性胰腺癌石蜡包埋块样本中有7例C-K-ras 12密码子发生了点突变,占36.8%,其中1例为两种突变形式(GAT、GTT);有9例Rb第21位外显子发生了点突变,占47.3%;而在胰腺癌恶性淋巴瘤、胰腺多形细胞癌和慢性胰腺炎样本中均未检出上述突变情况。结论:C-K-ras 12位密码子和Rb第21外显子点突变与胰腺癌的发生有一定的相关性。 Objectives : To study the correspondence of point mutation of oncogene C - K - ras and antioncogene Rb with the occurrence of pancreatic carcinomas. Methods: The point mutation of C - K - ras at 12 codon and Rb at 21th exon were detected by PCR -SSP in the araffin embedding samples of 19 cases of pancreatic carcinomas , 1 case of pancreatic lymphoma , 2 cases of pancreatic polymorphous cell carcinomas and 5 cases of cronic pancreatitis . Results: C - K -ras mutation at 12 codon were de- tected in 7 cases of the 19 pancreatic carcinoma samples ( 37% ) , one of which displayed two kinds of mutation ( GAT and GTT) ; Of 19 cases of pancreatic carcinomas,9 cases were detected with mutation of Rb 21th exon (47.4%) ; No mutation in C- K- ras 12th codon and Rb 12 exon were detected in cronic pancreatitis, pancreatic lymphorua and pancreatic polymorphous cell carcinomas samples. Conclusion : There is some correspondence of point mutation of oncogene C - K - ras and antioncogene Rb with the occurrence of pancreatic carcinomas.
