大剂量孕激素治疗对子宫内膜腺癌细胞凋亡和bcl-2、bax蛋白表达的影响

摘要 目的:探讨大剂量孕激素治疗对子宫内膜腺癌组织细胞凋亡及其调控基因bcl-2、bax蛋白表达的影响。方法:选取2000-2005年收住院手术治疗、资料完整的子宫内膜腺癌患者病理标本30例,应用原位细胞凋亡检测技术(TUNEL)检测孕激素治疗前后子宫内膜腺癌组织细胞凋亡指数,其中16例采用免疫组化S-P法检测bcl-2和bax蛋白表达。结果:孕激素治疗前后子宫内膜腺癌组织细胞凋亡指数、bcl-2和bax蛋白表达率分别为(6.00±5.68)和(27.67±2.41)、87.50%和75.00%、100.00%和93.75%。孕激素治疗后子宫内膜腺癌组织中细胞凋亡指数较治疗前明显增加(P〈0.01),其变化与bcl-2及bax蛋白表达无关。结论:大剂量孕激素治疗通过促进细胞分化、增加细胞凋亡而达到治疗子宫内膜腺癌的目的。 Objective: To investigate the influence of high dose progesterone on apoptosis and bcl-2, bax protein expression in patients with endometrial adenocarcinoma. Methods: Thirty patients with endometrial adenocarcinoma were received high dose progesterone treatment. Before and after treatment, the apoptotic cells were detected by the methods of terminal deoxynucleotidyl transferase nick end labeling, and 16 of them were examined bcl-2 and bax protein expression by immunohistochemistry. Results: The expression rate of apoptosis index and and bcl-2, bax before and after treatment were (6.00±5.68) and (27.67± 2.41), 87.50% and 75.00%, 100.00% and 93.75%, respectively. There were remarkable increment of apoptosis index in endometrial adenocarcinoma after high dose progesterone treatment (P 〈0.05), and their changes are not correlated to bcl-2 and bax(P 〈0.05). Conclusion: High dose progesterone treatment may play an important role in inducing cellular differentiation and apoptosis of endometrial adenocarcinoma tissues.
