农杆菌介导转基因小麦植株的获得和检测

摘要 利用建立的遗传转化体系,以携带Npt II筛选标记基因和胰蛋白酶抑制剂基因的LBA4404/pRok2(农杆菌/质粒)对山农995604小麦愈伤组织进行遗传转化,获得了三株含有胰蛋白酶抑制剂基因的转基因小麦。通过PCR扩增和Southern杂交检测,证明了外源基因已整合到了三株转基因小麦基因组中。 Using the system of transformation,shannong995604 were transformed with A.tumefaciens LBA4404 containing Npt II and CpTl.As a result transgenic wheat plants were obtained,and they were proved that foreign DNA have been intergrated into their wheat genome by using PCR amplification and Southern blotting analysis on the three transgenic wheat plants.
