γ-生育三烯酚对人结肠癌HT-29细胞中DNA损伤作用的研究 被引量:6

摘要 目的探讨γ-生育三烯酚对结肠癌细胞中DNA的损伤作用。方法γ-生育三烯酚与结肠癌HT-29细胞共培养,采用单细胞电泳法检测γ-生育三烯酚对HT-29细胞中DNA的损伤程度。结果γ-生育三烯酚对人结肠癌细胞株HT-29细胞中DNA分子有较大程度的损伤,且与处理的浓度存在一定的相关性。结论γ-生育三烯酚对结肠癌细胞具有很强的细胞毒作用,在γ-生育三烯酚作用下HT-29细胞的DNA断裂损伤增加,可能就是γ-生育三烯酚抗肿瘤效应的重要作用机制之一。 Objective To study the effect of γ-Tocotrienol on the DNA damage of colon cancer HT-29 cells. Methods The colon cancer HT-29 cells were cultured with γ-Tocotrienol in vitro. The DNA damage of colon cancer cells was determined by the single cell gel electrophoresis assay. Results γ-Tocotrienol could induce the DNA damage of colon cancer cell-lines HT-29 and show dose-response relationship. Conclusions The γ-Tocotrienol has great cytotoxic effect on colon cancer cells, the results indicating that the increasing of DNA damage in colon cancer may be a very important mechanism of γ-Tocotrienol anti-tumor effects.
