KA对原代培养胎鼠皮层神经元的毒性作用

摘要 目的探讨KA的神经毒性作用所引起神经元的损伤性改变。方法原代培养SD胎鼠皮层神经元,以不同浓度的KA分别作用12h、24h,用MTT检测法、显微镜形态学观察等方法进行研究。结果KA对神经元的毒性作用主要表现为神经元胞质空泡化,随着KA浓度的增加和作用时间的延长,其对神经元的细胞生长抑制率均增加(P<0.05)。结论KA对皮层神经元的毒性作用主要表现为以胞质空泡化为特征的类坏死样改变。 Objective To explore the change of neurons with injury caused by the neurotoxic effect of kainic acid(KA). Methods Primary cultural cortical neurons were treated by KA with different doses for 12h and 24h respectively, and their changes were estimated by MTT assay and microscopy. Result Cytoplasmic vacuole was found in neurons induced by KA and the cell growth inhibition ratio of neurons treated by KA increased with the concentration of KA increased and the acting time prolonged(P〈0.05). Conclusion The toxic effect of KA on neurons shown as the characteristic cytoplasmic vacuolization similar to the change of paranecrosis.
