摘要
以脱脂香菇粉为原料,进行了提纯条件的优化研究。结果表明,其提纯最佳条件为:温度80℃,0.5 mol/L的NaOH溶液浸提4 h,醇沉后经3%的活性炭脱色20 min,硼氢化钾-碱-Sevag法除蛋白,透析后超滤,最后经50%和80%乙醇分级沉淀得到香菇多糖两个组分Le-Ⅰ和Le-Ⅱ。产率分别为6.6%和3.8%。
With the degreased powder of lentinan as raw material, the optimal condition of the extraction and purification are studied. The results show that the optimal conditions are. at 80 ℃, the powder is extracted in 0.5 mol/L NaOH solution for 4 hours; being precipitate with alcohol and fade with activated carbon for 20 minutes; removed protein by Boron hydrogenation potassium-Alkali- Sevag; being ultrafiltered after dialysis. At last the two components Le-Ⅰ and Le-Ⅱ of Lentinan are get after the grade precipitation with 50 % and 80% ethanol, with rate of 6.6% and 3.8 % respectively.
