摘要 本文介绍一种研究快速反应动力学的实验技术——停流光谱法。此类仪器主要由液路和光路两个系统组成:前者可将反应物在数毫秒内混合,而后者的作用则是及时检测反应物的减少或生成物的增加。在实验中,可根据反应体系的吸收光谱选用适当的滤波片,根据化学反应的半寿期设定扫描时间;对于较长的扫描时间,可采用动态预热减小信号漂移。此外,本文还介绍了处理数据时应注意的事项,并列举了一个在生物学研究中的应用实例。 The aim of this paper was to introduce an experimental technique, the stopped-flow spectrophotometry, for the study of rapid reaction kinetics. The instrument of this kind was mainly composed of a liquid line and a light line. In the former, two reactants could be mixed in microseconds; and the decrease in reactants or the formation of the products was monitored by the latter. In a stopped-flow experiment, one should choose suitable light filter and time base. For a longer time base, dynamic warming up could effectively reduce signal drift. Some issues in data-processing were also discussed. Finally, application of the technique in biological study was shown with an example.
