摘要
利用所构建的减蛋综合征病毒六邻体蛋白基因真核表达质粒pcDNA2-hexon和减蛋综合征灭活油乳苗,观察了EDSV六邻体蛋白基因核酸疫苗对减蛋综合征灭活苗的免疫协同作用。结果显示,7日龄时应用核酸疫苗和减蛋综合征灭活油乳苗联合免疫的鸡群,在14~56日龄整个观察期内,胸腺T淋巴细胞对ConA、法氏囊B淋巴细胞对PMA的反应,均明显强于单纯油乳苗免疫组(其中,除在接种后第7天两组鸡的B淋巴细胞增殖OD570值差异不显著外,其余各检测时间两组间均表现为显著或极显著差异);特别是T淋巴细胞增殖情况在接种第14天后均表现为差异极显著。而且,HI抗体效价水平也持续性高于单纯灭活油乳苗免疫鸡群,并在免疫接种后35~49d时差异显著。说明EDSV六邻体蛋白基因核酸疫苗与灭活油乳苗联合免疫,可明显提高机体的细胞免疫和体液免疫水平,产生很好的免疫协同作用。
The immune efficacy of the combined inoculation of oil-adjuvant inactivated vaccine and pcDNA3--hexon gene eukaryotic expression plasmids (DNA vaccine) against egg drop syndrome virus (EDSV) was observed. The results showed that the proliferation response of the T lymphocytes from thymus and the B lymphocytes from bursa in chickens immunized with combined inoculation were strong than those of chickens immunized only with inactivated vaccine during all experiment period (P〈0.05 or P〈0.01, except the B lymphocytes from bursa in two group chickens on 7th day post-inoculation), especially the proliferation response of the T lymphocytes were all significantlt strong than those of chickens immunized only with inactivated vaccine after 14 day inoculation(P〈0.01). Meanwhile, the titers of HI antibody of chickens inoculated with both inactivated vaccine and DNA vaccine were higher than that of chickens inoculated only with inactivated vaccine during all experiment period, the difference of which was significant during 35--49 d post-immunization. It can be concluded that the combining use of oil-adjuvant inactivated vaccine and DNA vaccine could enhance highly the immune response and increase the immunity against EDSV.
