摘要
目的:探讨缺氧环境下缝隙连接蛋白(Cx40、Cx43)在mRNA水平上的变化,研究细胞缺氧和Cx40、Cx43表达变化在不稳定膀胱发病机理上的相关关系。方法:体外培养大鼠膀胱平滑肌细胞,建立细胞缺氧模型;在细胞缺氧1 h后收集细胞,运用逆转录-聚合酶链反应技术分析Cx40、Cx43。结果:缺氧1 h,Cx40、Cx43在mRNA水平上表达较正常氧对照组有明显升高(P<0.01)。结论:细胞缺氧会引起Cx40、Cx43在mRNA水平上表达的变化,这可能与不稳定膀胱的发生有密切联系。
Objective: To observe the connexin(Cx40, Cx43)changes under cell hypoxia in smooth muscle cells. Method: The smooth muscle cells were cultured in vitro. The mRNA levels of Cx40 and Cx43 were detected by RT-PCR one hour after cell hypoxia. Results: The mRNA levels of Cx40 and Cx43 in the one hour cell hypoxia group were higher than those in the cell normoxia group(P 〈 0.01). Conclusion: Cell hypoxia can cause the increase of mRNA of Cx40 and Cx43, and it might be contributed to the unstable bladder.
