不同单克隆抗体检测结肠癌P-糖蛋白表达的对比研究

摘要 目的:用单克隆抗体(JSB1、C219和C494)检测结肠癌组织中P-糖蛋白(P-gp)的表达。方法:应用免疫组化S-P法,通过JSB1、C219和C494分别检测96例结肠癌组织中P-gp表达的阳性率,分析与组织学类型、浸润程度、淋巴结转移和Dukes分期之间的关系以及结肠癌患者的生存情况。结果:96例结肠癌组织中用JSB1检测出P-gp阳性率为57.3%,C21933.3%,C494 95.8%,其与结肠癌的组织学类型、浸润程度、淋巴结转移和Dukes分期等均无明显相关;C494的检测结果与其它单克隆抗体检测结果的不同形式组合所显示出的结肠癌患者1、3和5年的生存率之间比有统计学意义(P<0.05)。结论:JSB1、C219和C494检测结肠癌组织中P-gp表达明显不同。 Objective: To study the monoclonal antibodies(JSB1, C219 and C494)for P-glycoprotein(P-gp)in colon carcinoma. Methods: JSB1, C219 and C494 were respectively used to detect the expression profiles of P-gp in 96 cases of colon carcinoma by the S-P method of immunohistochemistry, and the tissue type, invasion, lymph node metastasis, Dukes stages and patients' survival were analyzed. Results: The positive rates for P-gp detected by JSB1, C219 and C494 were 57.3%, 33.3% and 95.8% in 96 cases of colon carcinoma, respectively. The positive rate for P-gp expression detected by JSB1, C219 and C494 was not associated with the histologic grade, invasion, lymph node metastasis and Dukes stages in colon carcinoma. The 5-year survival rate in cases of positive expression was observed. Conclusion: JSB1, C219 and C494 are obviously different in detecting P-gp in colon carcinoma.
