琯溪蜜柚成年态茎段无菌系建立的影响因素

Establishment of aseptic strain from mature stem segments of Guanximiyou(Citrus grandis Osbeck)

摘要 以琯溪蜜柚田间成年树茎段为材料,研究外植体消毒方法和茎段不同成熟度等因素对无菌系建立的影响。试验结果表明,经70%酒精处理30 s、400 mg.L-1头孢拉定浸泡30 min的预处理后,用0.1%升汞溶液消毒5 min结合10%NaClO消毒30 min的方法,半木质化茎段的成功率最高,达83.3%;外植体切成5 cm长进行消毒以及培养基中添加活性炭有助于减轻外植体的褐变。 The effect of disinfection methods and developing stages of mature stem segments on the establishment of aseptic strain of Guanximiyou was studied. The results showed that after pre-treatment with 70% ethanol for 30s and 400mg·L^-1 cefotaxine for 30 min, the success rate of semi-hardened stem segments was highest, reaching 83.3 %, in the treatment of 0.1% HgC12 for 5 min and followed by the treatment of 10% NaCIO for 30 min. Cutting explants into segments of 5 cm long when disinfecting and adding activated carbon to the medium could reduce the browning rates of explants.
